This antibody recognizes a protein doublet of 20-22 kDa, identified as MART-1 (Melanoma Antigen Recognized by T cells 1) or Melan-A. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Seven other melanoma associated antigens recognized by autologous cytotoxic T cells include MAGE-1, MAGE-3, tyrosinase, gp100, gp75, BAGE-1, and GAGE-1. Subcellular fractionation shows that MART-1 is present in melanosomes and endoplasmic reticulum. This MAb labels melanomas and other tumors showing melanocytic differentiation. It is also a useful positive-marker for angiomyolipomas. Biotium Prostate Specific Antigen (PSA)(3E6), Biotin conjugate, 0.1mg/mL
Recognizes a single protein of 33-34 kDa, identified as the prostate specific antigen (PSA). This MAb is highly specific to PSA and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. PSA is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign prostate hyperplasia. Thyroperoxidase (TPO) is a 933 amino acid, type I transmembrane glycoprotein that plays a key role in thyroid gland function and autoimmunity. It is present as a dimer on the apical surface of thyroid follicular cells. TPO functions in the iodination of tyrosine residues in thyroglobulin and phenoxy-ester formation between pairs of iodinated tyrosines to generate the thyroid hormones, thyroxine and triiodothyronine. Mutations in this gene are associated with several disorders of thyroid hormonogenesis, including congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA. Malignant thyroid tumors exhibit an anomaly in TPO resulting in lower affinity for anti-TPO. This antibody may aid in the differentiation between benign and malignant thyroid tumors. Caldesmon HMW is the high molecular weight variant of Caldesmon. Two closely related variants of human caldesmon have been identified which are different in their electrophoretic mobility and cellular distribution. The h-caldesmon variant (120'150 kDa) is predominantly expressed in smooth muscle whereas l-caldesmon (70'80 kDa) is found in non- muscle tissue and cells. Neither of the two variants has been detected in skeletal muscle. This antibody recognizes only the 150 kDa variant (h-caldesmon) in Western blots of human aortic media extracts and is unreactive with fibroblast extracts from cultivated human foreskin. Caldesmon is a developmentally regulated protein involved in smooth muscle and non-muscle contraction. Recognizes protein of 192 kDa, which is identified as Clathrin Heavy Chain. Clathrin is composed of three heavy chains and three light chains, which associate non-covalently to form a triskelion structure. Clathrin heavy chain (HC) is composed of a terminal globular domain, a distal segment and a proximal segment containing a light chain-binding site. The proximal segment of the Clathrin HC protein is essential for interactions between Clathrin heavy chains and light chains, which result in the formation of the triskelion structure. Biotium Cytokeratin 10 (Suprabasal Epithelial Marker)(AE20), CF488A conjugate, 0.1mg/mL
This MAb recognizes a protein of 56. 5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Expression of CK10 is related to maturation of malignant keratinocytes, being preferentially detected in more-differentiated parts. Recognizes a protein of 40 kDa, identified as cytokeratin-19 (CK19), which is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. This MAb recognizes the peptide core of gastric mucin M1 (recently identified as Mucin 5AC). Its epitope is destroyed by beta-mercaptoethanol but not by periodate treatment. This mucin is present in primary ovarian mucinous cancer but usually absent in colorectal adenocarcinoma, thus showing an expression pattern opposite to MUC2. Together with a panel of antibodies, Anti-MUC5AC may be useful for differential identification of primary mucinous ovarian tumors from colon adenocarcinoma metastatic to the ovary. Reacts with a protein of 57 kDa, identified as the L1 protein of human papilloma virus type 16 (HPV-16). It is the major capsid protein of HPV-16. Infection with specific types of HPV has been associated with an increased risk of developing cervical neoplasia. HPV types 6 and 11 have been associated with relatively benign diseases such as genital warts but types 16 and 18 are strongly associated with cervical, vaginal, and vulvar malignancies. The antibody reacts very strongly with formalin-fixed, paraffin-embedded tissues containing HPV-16 or -33; very weak reactions were occasionally observed with biopsy specimens or smears containing HPV-6 or HPV-11. GP2 (glycoprotein 2), also known as ZAP75, is a 537 amino acid secreted protein. It is an integral membrane protein that is secreted from intracellular zymogen granules and associates with the plasma membrane via glycosylphosphatidylinositol (GPI) linkage. GP2 is cleaved and then released into the pancreatic duct along with exocrine secretions. GP2 binds pathogens such as enterobacteria, thereby playing an important role in the innate immune response. The C-terminus of this protein is related to the C-terminus of the protein encoded by the neighboring gene, uromodulin (UMOD). GP2 is also expressed on the apical plasma membrane of specialized microfold (M) cells among enterocytes and serves as a transcytotic receptor for mucosal antigens. S100A4 belongs to the S100 super-family of proteins containing 2 EF-hand calcium-binding domains. S100 genes include at least 25 members, including S100A1-S100A18, trichohyalin, filaggrin, repetin, S100P, and S100Z. S100A4 exerts its function via direct interaction with a number of proteins including P53, P63, non-muscle myosin IIA, α6β4 integrin, and liprin b1. This antibody reacts with Ig domain of CD47 protein. CD47, originally named integrin-associated protein (IAP), is a 50 kDa protein containing five membrane-spanning sequences and a short cytoplasmic tail. CD47 plays a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. It is important in memory formation and synaptic plasticity in the hippocampus. Recognizes a protein of 94 kDa, which is identified as the glucose-regulated protein 94 (grp94) and also tumor rejection antigen (gp96). Grp94 shows a high degree of sequence homology with the heat shock protein 90 (hsp90). This MAb is highly specific to grp94 and shows minimal cross-reaction with other members of the HSP90 family. Grp s are a class of proteins unresponsive to heat shock and are induced by glucose deprivation. The C-type lectin superfamily member CD72 is a cell surface negative regulator of B cell activation from the pro-B through the mature B cell stage. CD72 serves as a receptor for CD5. The human CD72 gene encodes a transmembrane glycoprotein that contains an immunoreceptor tyrosine-based inhibition motif (ITIM). Upon tyrosine phosphorylation, the CD72 ITIM recruits SH2-containing phosphatases such as SHP-1, resulting in downregulation of cell activation. CD72-/- mice contain hyperproliferative B cells. This MAb reacts with a 58 kDa protein identified as vimentin. It shows no cross-reaction with other closely related intermediate filament proteins (IFP s) such as desmin, keratin, neurofilament, and glial fibrillary acid protein.Anti-vimentin alone is of limited value as a diagnostic tool; however, when used in panels with other antibodies, it is useful for the sub-classification of a given tumor. Expression of vimentin, when used in conjunction with anti-keratin, is helpful when distinguishing melanomas from undifferentiated carcinomas and large cell lymphomas. All melanomas and Schwannomas react strongly with anti-vimentin. It labels a variety of mesenchymal cells, including melanocytes, lymphocytes, endothelial cells, and fibroblasts. Non-reactivity of anti-vimentin is often considered more useful than its positive reactivity, since there are a few tumors that do not contain vimentin, e.g. hepatoma and seminoma. Anti-vimentin is also useful as a tissue process control reagent.P
